The Root Cause

Why Energy Declines With Age

Aging isn't one thing โ€” it's a cascade of molecular failures. At the heart of most of them is one culprit: mitochondrial decline. Here's what's actually happening in your cells.

The Mitochondrial Aging Cascade

Every cell contains 1,000โ€“2,000 mitochondria โ€” the organelles that convert food into ATP, the universal energy currency. As we age, mitochondrial DNA accumulates damage, cristae structures deteriorate, and electron transport chain efficiency plummets. The result: less energy, more damaging reactive oxygen species (ROS), and a cellular environment that accelerates all other hallmarks of aging.

Peptides like MOTS-c, Humanin, and SS-31 work directly at the mitochondrial level โ€” either signaling from within the organelle or physically stabilizing its membrane architecture. This is why researchers consider them potentially transformative for aging biology.

The Journey

Anti-Aging Peptide Research Timeline

From the first thymic peptide isolation in 1974 to the 2025 confirmation of telomere elongation in human cells โ€” a half-century of science.

The Longevity Science Moment

We're living through an unprecedented moment in aging science. For the first time, researchers can identify specific molecular targets โ€” telomere length, mitochondrial function, NAD+ pools โ€” and design compounds to intervene at each point. The question is no longer whether aging can be slowed, but which interventions will prove safe and effective in humans.

The 2023 expansion of the Hallmarks of Aging framework gave researchers 12 distinct biological targets. Peptides already map onto at least 7 of them โ€” making this one of the most promising therapeutic classes for the coming decade.

Got Questions?

Frequently Asked Questions

Anti-aging peptides generate more hype โ€” and more confusion โ€” than almost any other field in health science. Here are honest, research-grounded answers.

MDPs are tiny proteins encoded directly in mitochondrial DNA โ€” not nuclear DNA. MOTS-c and Humanin are the two most studied examples. They act like internal distress signals: when a cell is metabolically stressed, MDPs are released to activate repair, reduce inflammation, and optimize energy production. Their circulating levels decline measurably with age โ€” and researchers believe restoring them may be one of the most direct ways to address the cellular energy crisis that drives aging.

All three stimulate growth hormone โ€” but through different mechanisms and durations. Sermorelin mimics the first 29 amino acids of natural GHRH, producing a short GH pulse close to natural physiology. CJC-1295 is a longer-acting GHRH analogue that extends GH pulse duration. Ipamorelin is a GH secretagogue (mimics ghrelin) that amplifies pulse height selectively without raising cortisol or prolactin. CJC-1295 + Ipamorelin is commonly combined to maximize GH pulses synergistically. Sermorelin is the only one FDA-approved (for pediatric use).

NAD+ (Nicotinamide Adenine Dinucleotide) is a coenzyme found in every living cell, required for energy production (ATP), DNA repair, and activation of sirtuins โ€” a family of longevity proteins. NAD+ levels decline roughly 50% between ages 40 and 70. Replenishing it via precursors like NMN or NR has shown benefits in multiple human RCTs: improved metabolic health, better muscle function, and vascular improvement. It's perhaps the best-evidenced longevity intervention after lifestyle measures.

It depends on the peptide. Sermorelin and Thymosin Alpha-1 have formal approvals and established safety profiles. NAD+ precursors have strong human RCT data. But most exciting peptides โ€” MOTS-c, Humanin, Epithalon โ€” are still in preclinical phases with no completed human clinical trials. The gap between exciting animal data and proven human benefit is wide. This is cutting-edge science, not proven medicine. For any therapeutic use, always work with a licensed physician.

The thymus gland is command central for your T-cell immune system. After puberty, it begins to shrink (a process called thymic involution) at approximately 3% per year โ€” progressively reducing the body's ability to generate new, effective T-cells. This is a major driver of immune senescence (the aging of the immune system). Thymosin Alpha-1 is a naturally occurring thymic peptide that can partially restore T-cell education and activation, countering this decline. It's approved in 35+ countries for immune restoration.

In 2023, Lopez-Otin et al. expanded the hallmarks of aging to 12 core biological processes: telomere attrition, genomic instability, epigenetic alterations, loss of proteostasis, disabled macroautophagy, deregulated nutrient sensing, mitochondrial dysfunction, cellular senescence, stem cell exhaustion, altered intercellular communication, chronic inflammation (inflammaging), and dysbiosis.

Peptides don't address all 12, but they map onto many: Epithalon targets telomeres, SS-31/MOTS-c target mitochondria, NAD+ links to nutrient sensing and DNA repair, and Thymosin Alpha-1 addresses immune senescence.

Legal & FDA Disclaimers

The excitement around anti-aging science is real โ€” but it demands equal parts rigor and responsibility.

Educational Purposes Only

All content on this page is strictly for educational and informational purposes. Nothing constitutes medical advice, a diagnosis, or a treatment recommendation. This information does not replace consultation with a qualified, licensed healthcare professional.

FDA Status โ€” Anti-Aging Peptides

Sermorelin is FDA-approved for pediatric GH deficiency only. Thymosin Alpha-1 is approved in 35+ countries but NOT for anti-aging in the US. NR (nicotinamide riboside) has GRAS status as a food ingredient. MOTS-c, Humanin, Epithalon, CJC-1295/Ipamorelin, and GHK-Cu (systemic) are NOT FDA-approved for any human therapeutic use.

Animal Lifespan Data โ‰  Human Results

Multiple compounds on this page have demonstrated lifespan extension in rodent models โ€” including MOTS-c, Humanin, and Epithalon. These findings are scientifically exciting but do not predict human outcomes. The history of longevity research is full of promising animal results that have not translated to humans. Preclinical data is hypothesis-generating, not proof.

Do Not Self-Administer Research Compounds

Many anti-aging peptides and compounds are sold online as 'research chemicals.' These are unregulated for human use, may be impure or mislabeled, and carry unknown health risks. Self-injection outside supervised medical settings is dangerous. House of Peptides does not endorse or facilitate this practice in any way.

Sermorelin (Geref) is FDA-approved for GH deficiency in children โ€” its off-label use in adults has not received FDA approval. Thymosin Alpha-1 is approved in select countries for specific immune indications. Nicotinamide riboside (NR) has GRAS status as a food ingredient; NMN’s regulatory status under the FDA is currently under review (FDA indicated in 2022 it may not qualify as a dietary supplement). All other peptides discussed โ€” including MOTS-c, Humanin, Epithalon, CJC-1295, Ipamorelin, SS-31, and GHK-Cu (systemic) โ€” are NOT approved for human therapeutic use.
